Bioassay-guided fractionation of CHCl3 extract from the fermentation broth of a sponge Mycale plumose-derived actinomycete Saccharopolyspora sp. nov., led to the isolation of two known prodigiosin analogs--metacycloprodigiosin (1) and undecylprodigiosin (2). These compounds exhibited significant cytotoxic activities against five cancer cell lines: P388, HL60, A-549, BEL-7402, and SPCA4. This is the first report on the significant cytotoxicity of metacycloprodigiosin (1) against human cancer cell lines.

The eukaryotic cell cycle is regulated by the sequential activation of cyclin-dependent kinases (CDKs). CDK activation is regulated by phosphorylation of the catalytic subunit, by binding to activating (cyclins) and inactivating subunits (cyclin-dependent kinase inhibitor). In this review, we will focus on the role of the cyclin-dependent kinase inhibitor p27 which has been recently the subject of extensive work. This negative regulator of cell growth indeed illustrates the pleiotropic biological effects of such molecules in both normal and cancer cells and the complexity of the regulatory mechanisms involved.

Being overweight increases the risk of developing many common diseases including type-2 diabetes mellitus, hypertension, coronary heart disease, gallstones and various cancers of the gastrointestinal and urogenital tracts. It can also cause or exacerbate osteoarthritis, breathlessness, heartburn, sleep apnoea, venous thromboembolism and psychological distress, particularly anxiety and depression. It makes anaesthesia and surgery more hazardous, and in pregnancy increases the risks associated with childbirth. Being overweight can also complicate day-to-day social functioning such as negotiating seats on public transport or purchasing clothes. In this article, we review the evidence that weight loss is beneficial and how this might be achieved using lifestyle changes, drug therapy, or surgery.

A recent analysis of epidemiological data from the Nordic Cancer Registries revealed that the prognosis of several groups of cancer patients in Denmark is inferior to that of the other countries. These differences have evolved during the last 20-30 years. The prognosis of cancer patients in the Nordic countries is reasonably good compared to other European countries and to the black community in the United States while the white community in the US has a better prognosis with regard to most of the cancer diseases. It is speculated that these differences reflect differences in resources allocated to cancer treatment.

The effect of a therapeutically administered high calorie diet in a severely malnourished patient is discussed in this case report. In patients with advanced head and neck cancer prolonged periods of malnutrition prior to admission are frequently encountered. This case report highlights the need to constantly monitor the electrolyte and vitamin levels during the early stages of instituting enteral or parenteral nutrition. By vigilant monitoring and a high index of suspicion re-feeding syndrome or severe hypophosphataemia and its associated complications can be avoided.

Smoking and drinking status did not affect results. These polymorphisms do not appear to be important gene modifiers in pancreatic cancer.

Sun exposure is the only major causative factor for skin cancer for which prevention is feasible. Both individual and community-based interventions have been effective in changing sun exposure knowledge and attitudes but generally have not been effective in changing behaviors. An integrative model of behavior change is described that has been successful in changing behavior across a wide range of health conditions. This model holds promise for developing a rational public health approach to skin cancer prevention based on sound behavioral science.

2-Phenylindole derivative 8, linked to aziridine by a hexamethylene spacer group, was synthesized and tested. It binds to the estrogen receptor with an RBA-value of 4.0 (estradiol = 100). In vitro, it shows a selective cytostatic activity in hormone-dependent MCF-7 breast cancer cells (41% T/C at 10(-6) M). In vivo, it exerts a strong estrogenic effect. The binding to the estrogen receptor is largely reversible.

From 1980 to 1988 more than 1,549 organ donors were harvested, 125 of them (8%) died from cancer--116 brain tumors and 9 donors with extracerebral cancer. In 3 donors suffering from extracerebral cancer 5 kidneys were transplanted. All transplants functioned well without tumor signs up to 21 months after grafting. However, because of tumor recurrence in nearly 50% of the cases described in the literature generally organs from donors suffering from extracerebral cancer should not transplanted if possible.

This article provides an overview of the classification of thyroid carcinoma into well-differentiated forms (papillary and follicular) as well as the rare Hurthle cell, medullary, and anaplastic forms. The characteristics, relative frequency, and mortality rates of each form of thyroid cancer are summarized. The roles of history, physical examination, blood tests, such as thyroid function test, thyroglobulin (htg), calcitonin and CEA, radionuclide and ultrasound imaging, and fine-needle aspirates are outlined for the diagnosis and management of thyroid cancer. An algorithmic diagram of the monitoring procedures for long-term management of various forms of thyroid cancer is presented.
