TITLE: History of electrophoretic methods.
PUBDATE: 19891001
Electrophoresis is the migration of electrically charged particles or ions in solutions due to an applied electric field. The ability to separate very similar substances including different proteins for analytical and preparative purposes has increased, especially since 1950, owing to the introduction of zone electrophoresis in paper and later in gels of polyacrylamide or agarose. After 1960, disc and displacement electrophoresis (isotachophoresis) and isoelectric focusing offered much increased resolution. Electrophoretic methods nowadays promote advances in biochemistry and molecular biology and will continue to be very important in science and for numerous applications in genetics, gene technology, sequencing of nucleic acids and proteins, studies of diseases and malfunctions including cancer, and in the identification of species and individuals, e.g., in forensic medicine.

A 31-year-old woman was diagnosed as having a malignant melanoma of the choroid in the right eye. Twenty months later her 7-year-old son was diagnosed as having retinoblastoma in the right eye. Both diagnoses were confirmed histopathologically after enucleation. A chromosomal study revealed normal karyotypes in the son. The family history was positive for cancer of the stomach. The implications of this rare association are discussed.

Some 40 accredited aftercare clinics are available for inpatient cancer aftercare, since 1981 also for immediately post-acute treatment courses. These clinics are used for both, the frequent physical treatment aftermath from oncological diseases in the otological or gynaecological field, and for psychic stabilization when psychosocial problems arise. Participation in autogeneous relaxation training, yoga, and/or psychotherapeutic services are found to entail improved disease coping. Concrete patient care expectancies primarily center around movement therapy, lymphatic drainage (in breast carcinoma), speech therapy (in laryngectomees), pain control, but are also directed at psychosocial care in terms of achieving" inner calmness and relaxation". Given the age distribution seen, increasing the quality of life takes priority over restoration of the working capacity.

PUBDATE: 19870301
The value of CEA and Ca 19-9 assays in the diagnosis and staging of lung cancer is examined. The particular sensitivity of CEA especially in extensive or metastasised tumours is demonstrated. The minimal sensitivity of Ca 19-9 is pointed out but also its absolute specificity in high risk control groups (smokers and BPCO). It is therefore concluded that Ca 19-9 might be used together with CEA in the diagnosis and staging of lung cancer.

PUBDATE: 19880101
The special radiobiological effects of low dose-rate radiation on tumors and normal tissue are in large part responsible for the effectiveness of brachytherapy. There is good reason to believe that improvement in local tumor control can be expected by the addition of chemotherapeutic agents, specifically designed radiation sensitizers, and differentiation-inducing agents, as well as by the manipulation of dose rates and dose distributions. This can be an important contribution to cancer management in the future and is an exciting area for further study.

PUBDATE: 19860501
We treated 17 patients with metastatic breast cancer with the four-drug combination of vincristine (0.5-1.0 mg), vinblastine (4 mg/m2), doxorubicin (40 mg/m2), and cyclophosphamide (400 mg/m2). The two vinca alkaloids appeared to be synergistic in causing acute, reversible neurotoxicity, manifested primarily as diffuse pains involving one or more of jaws, limbs (particularly joints), chest, and abdomen. The response rate of 70% was no higher than that noted previously with a regimen identical to this one except for the exclusion of vincristine. Further studies of combined vinca alkaloids are not recommended in the treatment of breast cancer.

Six hundred fifty-three biopsies were performed for clinically occult, mammographically detected breast abnormalities. One hundred forty-seven cancers (22.5%) were found. Eighty-nine of those cancers (60.5%) were noninvasive. None of the in situ lesions had involved axillary lymph nodes. Of the 58 invasive cancers, only six (10.3%) had metastases to axillary nodes. Fifty-four patients (36.7%) were treated by mastectomy while 93 patients (63.3%) were treated conservatively, 20 by biopsy only, and 73 by lumpectomy, axillary node dissection, and radiation therapy. Only four patients (0.7%) had significant complications.

PUBDATE: 19860601
The paper is concerned with the results of gamma-beam therapy of 120 patients with inoperable lung cancer using the fractionation of a common daily dose of 2-2.2 Gy into 2 equal fractions as a variant of super-fractionated irradiation used in clinical practice. The direct and short-term therapeutic results were improved: a complete tumor regression rate determined by x-ray was 26.7%, a 2-year survival rate 19.2%. In the group of patients who were irradiated by the commonly used methods, the same values were lower. Improved tolerance of therapy with fractionation of a common daily dose was noted.

Quantitation of circulating immune complexes (CIC) levels was established in patients with oral cancer and oral precancerous lesions. The levels were compared with that in normal controls and chronic chewers of betel quid with no signs of any disease. Both patients with oral cancer and oral precancerous lesions had elevated CIC when compared to both the control groups. The most interesting observations were (a) the CIC levels in the chewing controls were significantly raised when compared to normal controls; and (b) the CIC levels in the patients with premalignant lesions were elevated almost to the same levels as in the oral cancer patients.

PUBDATE: 19890401
An 18 year old girl presented with a leiomyosarcoma of the small bowel with widespread dissemination. Despite short term remissions after chemotherapy and surgery she died of the disease 10 months later. A history of prolonged exposure to danthron (an anthraquinone laxative) in childhood was obtained. Anthraquinones are known to be mutagenic and may be carcinogenic in experimental systems but danthron is not a proven carcinogen in man. The association of danthron and a rare bowel cancer in one case cannot prove a causative link. However, collection of further data seems advisable.

PUBDATE: 19880401
A proportionate mortality ratio (PMR) analysis of 1071 deaths in pulp and paper mill workers in New Hampshire during 1975-85 showed an increase in cancers of the digestive tract and lymphopoietic tissues. A similar analysis of deaths for 452 timber cutters and loggers failed to show excess PMRs for cancers of these sites. Despite methodological constraints, these results suggest that one or more of the exposures experienced by pulp and paper mill workers may pose a significant carcinogenic risk. More definitive epidemiological studies are required to determine particular high risk processes or specific aetiological agents.

131I labeled rabbit antibodies to the human epithelial intestinal antigen beta 1-MA was administered intravenously to nude mice together with human tumor grafts: colon cancer (CC), breast cancer, Ewing's sarcoma and hepatoma. Antibodies to beta 1-MA were selectively accumulated in CC only, excluding the other tumors. Iodinated nonspecific rabbit IgG in mice with CC were distributed like antibodies to beta 1-MA in the body of mice with control heterografts. A conclusion was made of the promising use of labeled antibodies to beta 1-MA in radioimmuno-scintigraphy of CC and its metastases.

A dramatic improvement in the survival of patients with testicular cancer has been witnessed in the 1970s. These advances are in the areas of diagnosis, staging, and monitoring of patients with this disease. The thrust of improvement includes the finding of sensitive and specific markers, the introduction of CDDP, and utilization of CT. Also, the finding of the efficacy of multidisciplinary treatment consisting of intensive chemotherapy and surgery has played a decisive role in the management of patients. This paper is devoted of a discussion of the basic sciences related to testicular cancer and the diagnosis and management of this tumor.

In women drawn from a randomised, population-based trial of mammography screening for breast cancer the overall actuarial survival and disease-free survival for up to 7 years of observation were similar in 94 patients with interval cancers and 178 control patients with cancer diagnosed independent of screening. The similarity was confirmed by multiple regression analysis. These results contradict the hypothesis that a high growth rate of the primary tumour is associated with a greater metastatic potential and do not support the concept that patients with interval cancers should receive more aggressive local or systemic treatment.

PUBDATE: 19870601
Eighty-three patients with advanced colorectal carcinoma were entered on a phase II trial of weekly iv aminothiadiazole (125 mg/m2) plus daily oral allopurinol (300 mg). There were five partial responses. Median survival of all patients on study was 36 weeks from entry (48 weeks for those without prior therapy and 34 weeks for those with previous chemotherapy). Toxicity was generally mild and consisted predominantly of stomatitis. In the dose given, aminothiadiazole has limited activity against metastatic colorectal cancer.
