Hormone treatment, radiotherapy and anti-cancer chemotherapy are often used to treat prostate cancer. However, there is no effective method of treating hormone-independent prostate cancer. In this study, we attempted to establish a new treatment method for hormone-independent prostate cancer. We developed a new recombinant adenovirus vector containing a suicide gene and controlled by a tissue specific promoter, and examined the usefulness of gene therapy for hormone-independence and PSA expression in prostate cancer. We have also examined the usefulness of gene therapy involving an adenovirus and various tumor suppressor genes for human prostate cancer cells, which are under trial in the United States.

The extraordinary specificity of immune responses mediated by T cells against individual syngeneic tumors has led to the concept that many tumor antigens are 'unique'. The recent isolation of several T-cell-recognized unique antigens from various murine and human tumors has shown that the antigenic peptides are caused by somatic mutations and, thus, are truly tumor-specific. The following review summarizes current knowledge about these mutant tumor-specific antigens and their possible role in the development and progression of cancer. It also discusses some functional differences between mutant tumor-specific and shared tumor antigens, which generally represent unaltered peptides, also present on some normal cells.

A specimen mount card has been described to orientate such specimens in three dimensions prior to radiological and pathological examination. Experience with 243 biopsies in this unit has shown it to be a useful aid in determining completeness of excision. It may also be used as a guide to further surgery when excision is incomplete and breast conservation is the treatment option of choice.

It presents as severe abdominal pain and tenderness, fever, and diarrhea associated with granulocytopenia. Gastrointestinal symptoms associated with chemotherapy for head and neck neoplasms include nausea and emesis, but not acute abdominal distress. We present, to our knowledge, the first case of neutropenic enterocolitis in a patient receiving cisplatin and fluorouracil chemotherapy for metastatic head and neck cancer.

In 1993 we started offering transrectal ultrasound-guided percutaneous cryosurgical ablation of the prostate (PCAP) as a treatment alternative for localized prostate cancer. Through 1994, 85 patients underwent PCAP and 53 had 3-6 month postoperative biopsies. A learning curve effect was observed: 46% of early patients had positive postoperative biopsies, versus only 9% of later ones. With repeat PCAP on the early patients, currently 87% of the biopsied patients are disease free. While longer follow-up is necessary, it appears that PCAP merits further investigation as a treatment alternative for localized prostate cancer.

This review summarizes the current status of this technique, which is in rapid evolution as new tracers and new imaging cameras become available.

The present review gives an overview of the clinical value of the tumour-suppressor gene, p53, and the CD44 cell adhesion molecule in colorectal cancer and the pitfalls encountered in the immunohistochemical detection of these proteins. Immunohistochemistry potentially forms a procedure applicable for routine diagnosis and prognostication. Therefore, p53 expression and the independent prognostic importance of CD44v6 expression is given particular emphasis, and other molecular events underlying colorectal carcinogenesis are only mentioned briefly.

Genetic alterations affecting p16(INK4a) and cyclin D1, proteins that govern phosphorylation of the retinoblastoma protein (RB) and control exit from the G1 phase of the cell cycle, are so frequent in human cancers that inactivation of this pathway may well be necessary for tumor development. Like the tumor suppressor protein p53, components of this "RB pathway," although not essential for the cell cycle per se, may participate in checkpoint functions that regulate homeostatic tissue renewal throughout life.

Experiments were addressed to investigate the mechanisms by which cholinergic stimulation is coupled to the enhancement of proliferation of small cell lung cancer cells H69. Muscarinic stimulation triggers the release of cytosolic Ca2+ and of inositol(1,4,5)trisphosphate with comparable time courses. The presence of alpha-bungarotoxin or the absence of Ca2+ in external medium suppresses enhancement of clonal growth induced by brief applications of nicotine. Here we suggest that Ca2+ mobilization represents a trigger for the enhancement of small cell lung cancer cell proliferation upon cholinergic stimulation.
