TITLE: Vitamin A, a useful biochemical modulator capable of preventing intestinal damage during methotrexate treatment.

The effects of vitamin A on methotrexate treatment of mice inoculated with L1210 leukaemia cells or sarcoma 180 cells were studied. The small intestine of tumour transplanted mice was severely damaged after methotrexate treatment. Coadministration of vitamin A with methotrexate protected the small intestine from methotrexate-induced damage. The protective effect of vitamin A was histologically and biochemically demonstrated. Further, coadministration of vitamin A did not inhibit in vivo antitumour activity of methotrexate. Thus, biochemical modulation used to prevent methotrexate-induced malabsorption by vitamin A coadministration will be of great use in methotrexate cancer chemotherapy.

Approximately 1/3 of T1 bladder cancers treated by endoscopic resection alone will progress. Prognostic factors are needed to help selecting appropriate treatment for these tumors. The purpose of the present investigation was to study the relation between p53 nuclear overexpression and disease progression. Tumors from 59 patients were studied by means of immunohistochemical nuclear staining. Forty tumors showed p53 nuclear overexpression while 19 tumors scored negative for mutations. We could not demonstrate any correlation between mutations of the p53 protein and tumor grading. p53 nuclear overexpression was not related to disease progression.

With the National Library of Medicine (NLM) menu-based interface, end users (physicians) averaged fewer steps per question, while with the BRS command-drive interface, intermediaries appeared to be more efficient. Cancer Information Service (CIS) searchers, who have more PDQ experience than end users or intermediaries, made greater use of command stacking to anticipate menu selections. Retrieval was more complete in the NLM system, where both the menus and predefined print formats assisted the searchers.

Restricting the evaluation of carcinogenic risk to genotoxicity or mutagenicity tests, as recently recommended by some researchers, is a hazardous operation, since the carcinogenic process involves a complex sequence of events in which no simple, necessary and sufficient condition is known for either the occurrence or non-occurrence of a cancer. None of the available tests exhibit a generalizable meaning, while none of the carcinogenic process phases should be neglected in prediction studies. In particular, ignoring promoter action and progression or proliferation, as well as targets specificity and other points, should lead to an underestimation of the actual risk.

The occurrence of testicular tumors in closely related family members is rare. We report a case of testicular cancers in a father and son. The father had pure seminoma, and his son developed a mixed nonseminomatous germ cell tumor. The peripheral blood lymphocytes in both patients were tested for 52 HLA specificities, and the patients were found to have six in common. The son was homozygous for HLA A24. The association between certain HLA antigens, other genetic factors, and testicular cancer requires further study to improve the care and counseling of patients and their families.

Preservation of the pelvic nerve plexus at the sites of uterosacral and cardinal ligaments was practiced during radical hysterectomy in 104 cases of cervical cancer (study group). Conventional radical hysterectomy without preservation of nerve plexus was performed in 100 cases (control group). The amount of residual urine was measured postoperatively to evaluate the recovery of function of urination. The average time of recovery of normal urination was 18.25 +/- 1.93 days for the study group and 24.05 +/- 1.68 days for the control group (P less than 0.01). There was no difference of 5- and 10-year survival rates between these two groups (P greater than 0.05).

It has been shown repeatedly that behaviour therapy fulfils a useful prophylactic function in the prevention of cancer and coronary heart disease. The present study investigates the effects of orthodox psychoanalytic treatment on the eventual probability of death from cancer and coronary heart disease in probands treated by psychoanalysis for over two years, probands who broke off psychoanalytic treatment after less than two years, and various control groups. Psychoanalysis is shown to have a significantly negative effect on survival after seven years follow-up.

Folate deficiency significantly increases uracil content and chromosome breaks (as measured by micronucleated cells) in human leukocyte DNA. Folate supplementation reduces both the uracil content of DNA and the frequency of micronucleated cells, indicating that uracil misincorporation may play a causative role in folate deficiency-induced chromosome breaks. A calculation is presented to explain how the levels of uracil found in DNA could cause chromosome breaks. Based on this calculation, the frequency of uracil repair events that might result in double-strand DNA breaks increases by 1752-fold. These results are consistent with clinical and epidemiological evidence linking folate deficiency to DNA damage and cancer.

This paper describes the findings of a case-controlled study of 100 patients with primary liver cell carcinoma, sex- and age-matched with another 100 patients seen at the orthopaedic clinic of the University College Hospital, Ibadan. Contrary to widespread belief, the relative risks of cigarette smoking and alcohol consumption were not found to be statistically significant. The implication of this on the possible aetiology of primary liver cancer is discussed.
