TITLE: Vitamin A, a useful biochemical modulator capable of preventing intestinal damage during methotrexate treatment.
PUBDATE: 19930801
The effects of vitamin A on methotrexate treatment of mice inoculated with L1210 leukaemia cells or sarcoma 180 cells were studied. The small intestine of tumour transplanted mice was severely damaged after methotrexate treatment. Coadministration of vitamin A with methotrexate protected the small intestine from methotrexate-induced damage. The protective effect of vitamin A was histologically and biochemically demonstrated. Further, coadministration of vitamin A did not inhibit in vivo antitumour activity of methotrexate. Thus, biochemical modulation used to prevent methotrexate-induced malabsorption by vitamin A coadministration will be of great use in methotrexate cancer chemotherapy.

PUBDATE: 20031101
Cancer is frequently complicated by thrombosis, which may even be the first sign of an unrecognized cancer. There are several risk factors for cancer-associated thrombosis, such as type of malignancy, the presence of metastatic disease, and the use of chemotherapy. In most patients with cancer, a procoagulant state can be identified, ranging from subclinical laboratory abnormalities to full-blown disseminated intravascular coagulation. A specific entity is the occurrence of thrombotic microangiopathy that is specifi cally related to (high-dose) chemotherapy and radiotherapy. The pathogenetic pathways that play a role in the cancer-associated coagulopathy have been identified in recent years.

Multiple prognostic factors for outcome in papillary thyroid carcinoma are identified. For a cohort of 1500 consecutive patients followed an average of 16 years, survival rates for tumor recurrence and cause-specific mortality are described. Risk groups derived from novel prognostic scoring or staging systems and the role of DNA ploidy testing are discussed specifically. Lastly, controversies are reviewed regarding the extent of primary surgical resection and the efficacy of postoperative radioiodine remnant ablation in papillary thyroid cancer.

PUBDATE: 19951201
Approximately 1/3 of T1 bladder cancers treated by endoscopic resection alone will progress. Prognostic factors are needed to help selecting appropriate treatment for these tumors. The purpose of the present investigation was to study the relation between p53 nuclear overexpression and disease progression. Tumors from 59 patients were studied by means of immunohistochemical nuclear staining. Forty tumors showed p53 nuclear overexpression while 19 tumors scored negative for mutations. We could not demonstrate any correlation between mutations of the p53 protein and tumor grading. p53 nuclear overexpression was not related to disease progression.

The three standard projections relative to mammographies of 40 patients with breast cancer pre-operatively classified as T1-T2 were analyzed by computerized morphometric elaborations. This method si able to better define view of images, permetting pre-operative connection of staging, with an increment of 30% if confronted with histologic TNM (pTNM). The utilization of method is discussed to evaluate the best way for the local definition of tumor extension, permitting to use a correctly conservative surgery (quadrantectomy, tumorectomy) in a greater number of patients.

TITLE: User interactions with the PDQ cancer information system.
PUBDATE: 19920101
Searches by end users and intermediaries on the online PDQ (Physician Data Query) cancer information system were observed. With the National Library of Medicine (NLM) menu-based interface, end users (physicians) averaged fewer steps per question, while with the BRS command-drive interface, intermediaries appeared to be more efficient. Cancer Information Service (CIS) searchers, who have more PDQ experience than end users or intermediaries, made greater use of command stacking to anticipate menu selections. Retrieval was more complete in the NLM system, where both the menus and predefined print formats assisted the searchers.

QUESTION: A 35-year-old woman attending my clinic in the 20th week of pregnancy had a lump in her breast. Open biopsy revealed ductal carcinoma. Radical mastectomy with axillary lymph node biopsy showed three out of 10 nodes positive. What are the short- and long-term implications of chemotherapy during pregnancy? ANSWER: Patients with breast cancer during pregnancy should be treated using the same criteria as their nonpregnant counterparts, with the modifications required by pregnancy. The benefits of the diagnostic workup and use of chemotherapy, radiotherapy, and surgery have to be weighed carefully against their risk to the unborn baby.

A 70-year-old man was admitted with a nodule adjacent to a cystic lesion in the right lower lung field. Diagnosis of lung cancer was made by transbronchial biopsy. A series of chest radiographs revealed that a cystic lesion had presented 8 years before, and that a nodule appeared on the hilar side of the cyst 5 years later. The cyst may have been formed by the check-valve mechanism due to the lung cancer.

TITLE: [Detection of PSA mRNA in prostate cancer patients' blood]
PUBDATE: 20011001
A sensitive technique using reverse transcriptase-polymerase chain reaction (RT-PCR) for PSA mRNA has been used to detect circulating tumor cells in the peripheral blood of prostate cancer patients. We evaluated the clinical utility of this method for staging and monitoring for prostate cancer. The number of patients who were RT-PCR positive was increased in higher clinical stages. This technique may provide useful information for treating patients with prostate cancer, especially candidates for radical prostatectomy. Confirming the value of this modality as a prognostic factor will require further study.

TITLE: PTEN and myotubularin phosphatases: from 3-phosphoinositide dephosphorylation to disease.
PUBDATE: 20021201
The phosphatase and tensin homolog deleted on chromosome ten (PTEN) and myotubularin (MTM1) represent subfamilies of protein tyrosine phosphatases whose principal physiological substrates are D3-phosphorylated inositol phospholipids. As lipid phosphatases, PTEN- and MTM1-related (MTMR) proteins dephosphorylate the products of phosphoinositide 3-kinases and antagonize downstream effectors that utilize 3-phosphoinositides as ligands for protein targeting domains or allosteric activation. Here, we describe the cellular mechanisms of PTEN and MTMR function and their role in the etiology of cancer and other human diseases.

TITLE: The survival kinases Akt and Pim as potential pharmacological targets.
PUBDATE: 20051001
The Akt and Pim kinases are cytoplasmic serine/threonine kinases that control programmed cell death by phosphorylating substrates that regulate both apoptosis and cellular metabolism. The PI3K-dependent activation of the Akt kinases and the JAK/STAT-dependent induction of the Pim kinases are examples of partially overlapping survival kinase pathways. Pharmacological manipulation of such kinases could have a major impact on the treatment of a wide variety of human diseases including cancer, inflammatory disorders, and ischemic diseases.

TITLE: Beware of a new carcinogenic risk: promotion of promoters.
PUBDATE: 19951001
Restricting the evaluation of carcinogenic risk to genotoxicity or mutagenicity tests, as recently recommended by some researchers, is a hazardous operation, since the carcinogenic process involves a complex sequence of events in which no simple, necessary and sufficient condition is known for either the occurrence or non-occurrence of a cancer. None of the available tests exhibit a generalizable meaning, while none of the carcinogenic process phases should be neglected in prediction studies. In particular, ignoring promoter action and progression or proliferation, as well as targets specificity and other points, should lead to an underestimation of the actual risk.

TITLE: A rare case of penile metastasis of testicular cancer presented with priapism.
PUBDATE: 20050901
Priapism is thought as a condition of penile erection that persists beyond or is unrelated to sexual stimulation. Commonly two different entities of priapism are known, one is low-flow priapism and the other is high-flow priapism. It is important to distinguish these two conditions for the subsequent different treatments. We report a rare case of an indistinguishable priapism caused by penile metastasis of testicular cancer.
