TITLE: Recent trends in surgical treatment of localized prostate cancer.
PUBDATE: 20050901
Prostate cancer and its various forms of treatment remain a source of significant controversy and morbidity despite recent advances. In response, there is an increasing trend toward the development of treatments aimed at cancer prevention and at maximizing the preservation of function without sacrificing cancer control. This article reviews the current prostate cancer literature and reports on improvements in existing surgical treatments and developing technologies aimed toward achieving these goals. Specific therapies addressed include improvements in surgical techniques, laparoscopy, robotics, cryosurgical and thermal ablation, and high-intensity focused ultrasound.

We herein report about 2 heart transplant patients undergoing surgical resection for esophageal cancer. Both were long-term survivors after orthotopic heart transplantation for dilated cardiomyopathy. One patient underwent a transthoracic esophagectomy and gastric pull-up for an early squamous cell carcinoma of the infracarinal esophagus. The second patient underwent a resection of the cervical esophagus and interposition of a free jejunal segment after neoadjuvant radiochemotherapy for a locally advanced squamous cell carcinoma in the cervical esophagus.

TITLE: Role of integrins in cell invasion and migration.
PUBDATE: 20020201
As cancer cells undergo metastasis--invasion and migration of a new tissue--they penetrate and attach to the target tissue's basal matrix. This allows the cancer cell to pull itself forward into the tissue. The attachment is mediated by cell-surface receptors known as integrins, which bind to components of the extracellular matrix. Integrins are crucial for cell invasion and migration, not only for physically tethering cells to the matrix, but also for sending and receiving molecular signals that regulate these processes.

PUBDATE: 20031101
Arachidonic acid metabolites including prostaglandins (PG) participate in many biological processes and play regulatory functions. The significant role of PG in physiological and pathological process is well known and described by many authors. Prostaglandins were demonstrated to have local hormone properties and often exert an antagonist effect. Despite years of research the role of PG in cancer diseases development is still poorly understood. The article is focused on the influence of arachidonic acid metabolites and its analogs on tumor growth and metastases formation.

There were presented the late results of surgical treatment in 425 patients, suffering upper ampullar and rectosygmoid rectal cancers, to whom transabdominal rectal resection with the duplicature anastomosis formation in 1989--2002 yrs was done. In 93.4% of observations, according to histological investigation data, adenocarcinoma was diagnosed. Concurrent diseases were noted in 226 (53.2%) of patients. The postoperative complications rate was 13.2%, postoperative mortality--2.6%. Late result of treatment was fair.

PUBDATE: 20050401
Basal cell carcinoma is the most frequent cancer in humans. Several clinical types are distinguished. They are bound to distinct evolutive prognosis. The surgical excision is the treatment of choice which is rarely followed by recurrence. However, when the lesion is superficial and non aggressive and when the body site is adequate, topical applications of imiquimod can provoke the neoplastic regression. This type of immunotherapy brings 70 to 90% complete remission. A medical follow-up of the treated site is mandatory for a couple of years.

TITLE: Lysosomal cysteine proteases: structure, function and inhibition of cathepsins.
PUBDATE: 20051201
Lysosomal cysteine proteases, a subgroup of the cathepsin family, are critical for normal cellular functions such as general protein turnover, antigen processing and bone remodeling. In the past decade, the number of identified human cathepsins has more than doubled and their known role in several pathologies has expanded rapidly. Increased understanding of the structure and mechanism of this class of enzymes has brought on a new fervor in the design of small molecule inhibitors with the hope of producing specific, therapeutic drugs for diseases such as arthritis, allergy, multiple sclerosis, atherosclerosis, Alzheimer's disease and cancer.

TITLE: Developing an Oncology Portfolio of Anticancer Drugs: The Experience of one Pharmaceutical Company.
PUBDATE: 20051201
For a company or institute that wishes to develop new anti-cancer agents, it is necessary to establish a portfolio of agents to reduce the risk of failure. Success rates for developing new agents are low and therefore different biological effect areas should be explored to ensure that at least one agent targeting cancer or a specific histological sub-type of cancer is effectively developed. This paper describes how one pharmaceutical company has developed a range of different agents with the ultimate aim of developing as many of these as is technically feasible into useful new medicines for the treatment of cancer.

Early detection and prevention of gastrointestinal cancer involves screening and surveillance. The efficacy of early detection tests should be subjected to well-designed studies. Clinical recommendation should be made after evaluation of new therapeutic approaches that will be based on our understanding of the molecular and cellular biology and the genetics of gastrointestinal premalignancy and cancer. Chemoprotection and alterations in diet and environment are areas that offer much promise and require clinical evaluation.

TITLE: HGF/SF-Met signaling in tumor progression.
PUBDATE: 20050101
Tumor progression is a multi-step process that requires a sequential selection of specific malignant phenotypes. Met activation may induce different phenotypes depending on tumor stage: inducing proliferation and angiogenesis in primary tumors, stimulating motility to form micrometastases, and regaining the proliferation phenotype to form overt metastases. To study how HGF/SF-induced proliferative phenotypes switch to the invasive phenotype is important for understanding the mechanism of tumor progression and will provide an attractive target for cancer intervention and therapy.

We investigated breast cancer incidence in migrants in the Netherlands in 1988-1998. The standardised incidence ratio for breast cancer in Northwest-Netherlands was statistically significantly reduced for women born in Surinam (0.56), Turkey (0.29) and Morocco (0.22). The proportion of women with advanced stages (III and IV) did not differ significantly between migrants and women born in the Netherlands.

Eight randomized controlled trials of mammography screening have been conducted to date. In addition to evaluating the efficacy of screening with an experimental design, the trials provided investigators with access to information about breast cancers much earlier in their development than had previously been available. The trials of mammographic screening provide conclusive evidence that the policy of offering screening is associated with a significant and substantial reduction in breast cancer mortality.

TITLE: The RAF proteins take centre stage.
PUBDATE: 20041101
Since their discovery over 20 years ago, the RAF proteins have been intensely studied. For most of that time, the focus of the field has been the C-RAF isoform and its role as an effector of the RAS proteins. However, a report that implicates B-RAF in human cancer has highlighted the importance of all members of this protein kinase family and recent studies have uncovered intriguing new data relating to their complex regulation and biological functions.
