TITLE: Isolation and identification of a cytotoxic principle from Chrysosplenium grayanum Maxim. (Saxifragaceae) and its antitumor activities.
PUBDATE: 19921201
A cytotoxic principle was newly isolated from Chrysosplenium grayanum Maxim. (Saxifragaceae) and identified as beta-peltoboykinolic acid (1) on the basis of spectral data. Cytotoxicity of compound 1 was tested against various human cancer cell lines in vitro, and antitumor effect of this compound was demonstrated on Meth.A mouse fibrosarcoma. The experiment of combined treatment with compound 1, mitomycin C, and OK-432 resulted in enhancing the antitumor effect against B16-BL6 mouse melanoma in C57BL/6 mice.

TITLE: Inhibition of human colon cancer growth by antibody-directed human LAK cells in SCID mice.
PUBDATE: 19930301
Advanced human colon cancer does not respond to lymphokine-activated killer (LAK) cells. In order to direct cytotoxic cells to the tumor, human LAK cells linked with antibodies to a tumor cell surface antigen were tested with established hepatic metastases in severe combined immunodeficient (SCID) mice. These cells had increased uptake into the tumor and suppression of tumor growth as compared with LAK cells alone, thereby improving the survival of tumor-bearing mice. Thus, tumor growth can be inhibited by targeted LAK cells, and SCID mice can be used to test the antitumor properties of human effector cells.

In patients with advanced breast cancer the incidence of diabetes insipidus is between 0.1% and 0.9%. Satisfactory symptomatic relief can be obtained with Desmopressin-acetate. In the presence of this symptom complex magnetic resonance imaging (MRI) can aid in the detection of metastases to the posterior pituitary. By the use of magnetic resonance imaging, the incidence for and implementation of local radiotherapy can be firmly grounded.

The effects of smoking on female reproductive life are clear, with a longer time-lapse necessary for conception, alterations in hormonal and placental functions and an increased risk of extra-uterine pregnancy, prematurity and low birth-weight. The effects of passive smoking among children, irrespective of source, are linked to an increased risk of sudden infants death syndrome, as well as a higher frequency of otitis and respiratory ailments. Some authors have also found an increased risk of cancer in children and a negative impact on physical and psychological development.

Rheumatic manifestations of malignancy include a wide spectrum of osteoarticular, muscular, glandular, endocrinologic, and systemic features, posing a therapeutic challenge. The clinician should be aware that Sjögren's syndrome, polymyositisdermatomyositis, rheumatoid and rheumatoid-like arthritis, polymyalgia rheumatica and temporal arteritis, and diverse osteomuscular conditions may be the immunopathogenic features of a neoplasm, the direct consequence of osteomuscular tumors, the effect of tumor-associated hormones, or the consequence of cancer therapy. The principal articles that have appeared in the past year on these associations are discussed. We also review the association of x-ray irradiation and cancer in patients with ankylosing spondylitis.

TITLE: A review and biological risk assessment of sodium saccharin.
PUBDATE: 19920601
Dietary sodium saccharin is associated with bladder tumors when fed at high levels to the male rat. Under these conditions urinary pH, sodium concentration, and volume are elevated and proliferative changes are present in the urothelium. Extensive epidemiological studies have shown that saccharin does not increase the risk of bladder cancer in humans and laboratory investigations have shown that sodium saccharin is not mutagenic and does not bind to DNA. Recent research indicates that the urothelium in male rats is damaged under conditions of high urinary pH and sodium levels by a mechanism that involves alpha 2u-globulin and possibly silicate crystalluria. These studies and their implications for human health risk are reviewed.

PUBDATE: 19910701
Pancreatic cancer is the fifth leading cause of death from malignant disease in Western society. Apart from the fortunate few patients who present with a resectable small pancreatic adenocarcinoma, conventional treatment offers no hope of cure and has little palliative value. Over the past two decades major steps have been made in our understanding of the biology of pancreatic growth and neoplasia. This review sets out to explore these advances, firstly in the regulation of normal pancreatic growth, and secondly the mechanism which may be involved in malignant change of the exocrine pancreas. From an understanding of this new biology, new treatment strategies may be possible for patients with pancreatic cancer.

PUBDATE: 19910501
Though many previous reports suggest the clinical significance of adjuvant chemotherapy in gastric cancer, they are not always confirmed by the statistics. Treatment failures could be attributed to the minor benefit of chemotherapy for advanced cases, too much tumor burdens after surgery relative to the antitumor effect of chemotherapy, and statistically inadequate operation of clinical study. Improvement would be expected in the near future by carrying out a well designed control study with incorporation of new drugs such as CDDP or etoposide, or powerful regimen with MTX and 5 FU, or 5 FU and Leucovorin.

PUBDATE: 19940301
The development of risk assessment in occupational and environmental cancer control is outlined, with emphasis on the differing assumptions of Doll and Selikoff. These differences are explained by philosophic perspectives from which closed and open system approaches to assessment were respectively chosen. Open system, with negative entropy and progressive organization, are characteristic of living systems and, heuristically, should be characteristic of risk assessment models. The abuse of either approach occurs when the "habit of truth" is overcome by consciously contrived confusion, a betrayal of society by les clercs.

Breast examination for early cancer detection in high risk patients are being performed even more frequently in our country and in other ones. Wide availability of mammosonography facilitates such management. X-ray mammography in view of irradiation is less frequently used. An attempt has been undertaken at carrying out breast examinations of a selected Pałuski area. The analysis included 180 women who after carrying out comprehensive breast examination and obtaining of history data were qualified for the group of high risk for breast cancer.

Community health care and, more specifically, home health care are major developments over recent years. In many European countries there has been serious debate on the necessity of home care, the importance of this type of health care for the patient and family, and those factors influencing patient choice with regard to health care at home or in hospital. This question of choice between home and hospital care forms the basis of this piece of research, the aim of which is to examine the biosocial factors that influence the decision of cancer patients in Greece to choose health care at home or in hospital.

Two oral anticancer agents, cyclophosphamide and UFT (mixture of futraful and uracil), were combined with hormone therapy for 42 patients with prostatic cancer. Although UFT could be taken for long periods, cyclophosphamide arose many problems such as the bone marrow suppression and gastrointestinal disturbances. The median length of the days they could take cyclophosphamide was sixty days. The survival rate of patients who were newly diagnosed and treated with this regimen was better than patients received hormone therapy only, and it suggests the possibility of the combined chemotherapy to inhibit the progression frequently observed after conventional hormone therapy.

TITLE: Relationship between DNA methylation and gene expression of the HOXB gene cluster in small cell lung cancers.
PUBDATE: 19960201
The expression pattern of the HOXB gene cluster in four xenografted small-cell lung cancers was compared to the methylation of the DNA in the corresponding genomic regions. In 90% (17/19) of the studied cases, the expressed genes were in methylated regions whereas 70% (12/17) of the unexpressed genes were in unmethylated regions. This specific behavior could correspond to a particular gene expression regulation mechanism of the HOX gene network. Since some genes (HOXB2, HOXB4, HOXB7) were always inactive when unmethylated, this unexpected relationship might indicate their key function(s) in the HOX gene network.
