TITLE: Cell cycle checkpoints and their inactivation in human cancer.
PUBDATE: 20001001
Checkpoints are mechanisms that regulate progression through the cell cycle insuring that each step takes place only once and in the right sequence. Mutations of checkpoint proteins are frequent in all types of cancer as defects in cell cycle control can lead to genetic instability. This review will focus on three major areas of cell cycle transition control, with particular attention to the alterations found in human cancer. These areas include the G1/S transition, where most cancer-related defects occur, the G2/M checkpoint and its activation in response to DNA damage, and the spindle checkpoint.

TITLE: [Middle ear carcinoma]
PUBDATE: 19980401
Middle-ear cancer represents 5 to 10% of all ear neoplasms. Risk factors include chronic suppurative pathology of the middle ear and prior irradiation. Most malignant tumors are squamous-cell carcinomas. We report the case of a 51-year-old woman with this process. The ideal treatment is mastoidectomy or pterosectomy followed by radiotherapy. We made a bibliographic review of the clinical manifestations, diagnosis, treatment, and prognosis of middle-ear cancer. Five-year survival rates range from 25 to 50% in patients who undergo surgery and radiotherapy.

TITLE: [The treatment of the initial stage of cervical cancer]
PUBDATE: 20000301
The analysis of the author's own experience and of relevant published literature suggest that it is expedient to further elaborate methods for treatment of incipient forms of uterine cervix cancer aimed to preserve the genital organs in women, especially in those at childbearing age. Worthy of mention in this respect is the progress made by the Kiev City Oncological Centre. In choosing a treatment option, it is necessary that etiopathogenic risk factors for development of cancer of the uterine cervix by analyzed and taken account of.

Their structures were elucidated by spectroscopic methods. Compounds 1, 2, 3, and 5 showed low antitumor activity against four cancer cell lines.

Cannabigerol (3) exhibited the highest growth-inhibitory activity against the cancer cell lines.

TITLE: Dietary phytoestrogens in the prevention of long-term postmenopausal diseases.
PUBDATE: 19991001
Phytoestrogens (PEs) are natural compounds, with a biological activity like estrogen, which comprise isoflavones, lignans and coumestans. A traditional Asiatic phytoestrogen-rich diet is associated with a lower incidence of breast cancer and postmenopausal illness, and much evidence indicates that PEs prevent bone resorption, increase bone density and reduce cholesterol. The estrogenic effects of phytoestrogens can be useful in preventing postmenopausal osteoporosis and cardiovascular disease.

PUBDATE: 19990801
People with HIV/AIDS are vulnerable to primary central nervous system lymphoma (PCNSL), a type of brain cancer. Treatment choices are limited, as chemotherapy tends to further weaken the immune system, and radiation does not seem to have a major impact on survival. One promising treatment approach used AZT, Ganciclovir, and IL-2. A five-person clinical trial was conducted using this drug combination, and radiation if necessary. The subjects generally responded well to therapy. However, further research is needed to verify the findings and investigate optimal dosing.

The omega-conopeptide, ziconotide, is an N-type calcium-channel blocker that has been shown to produce antinociception in animals using formalin and hot-plate tests. Initial reports of intrathecal administration of ziconotide in cancer and AIDS patients whose pain was unrelieved with opioids demonstrated analgesic efficacy. Although adverse effects were reported, these appeared to be easily managed through dose reduction or symptomatic treatment. This clinical report describes the experiences of three patients with serious adverse effects associated with intrathecal ziconotide.

PUBDATE: 19990601
Pharmacoprevention of cancer is an attractive concept. For breast cancer, several trials were set up some years ago. The largest one was stopped before its planned date of closure and demonstrated an early appearing reduction in breast cancer occurrence; in contrast, the other two trials did not find such a protective effect. The decision is all the more difficult because of tamoxifen side effects, including carcinogenicity. Currently, no definite conclusion can be drawn.

A committee comprising hospital and community-based urology and oncology nurses and social workers planned, organized, implemented, and evaluated an educational public seminar on prostate cancer (PC). Data relating to satisfaction with the seminar, reasons for attendance, perceived needs for further support/education, and demographics were collected using a feedback questionnaire. Results suggested a need for education and a significant interest in support groups and further educational forums for patients with PC or symptoms and their caregivers.

The Pap smear unquestionably is a successful screening test for cervical cancer. However, recent advances in technology have raised questions regarding whether the conventional Pap smear is still the standard of care. This article relates issues of screening and cost-effectiveness to the state of the art in thin layer preparations, cytology automation, human papillomavirus screening, human papillomavirus vaccines, and other cervical screening adjuncts. Perhaps nowhere in medicine is clinical decision making being more strongly influenced by market and other external forces than in cervical cytopathology.

PUBDATE: 19990901
A general review of the results and technique of intraarterial chemotherapy in cases of breast cancer is given. The remission rate for untreated primary tumors is nearly 100%, for completely treated local relapses approximately 70%. The complication rate is quite low using an intraarterial well-tolerated cytostatic agent such as mitoxantrone. However, thromboembolism of the vertebral artery facing the internal mammary artery may occur. The indication for intraarterial chemotherapy should always be set by an interdisciplinary board.

PUBDATE: 20001001
SYMPTOMS: A 40-year-old woman presented with an arthritis of the knees and ankles, digital clubbing and thrombocytopenia. EXAMINATIONS: Blood tests proved an inflammatory reaction. Clinical findings were the above mentioned oligoarthritis and a periostitis of the long extremity bones. Chest X-ray showed a lung tumor. THERAPY: After surgical treatment of the lung cancer both arthritis and thrombocytopenia normalized. CONCLUSION: Hypertrophic osteoarthropathy is characterized by the combination of clubbing, arthropathy and periostitis. This paraneoplasia may be the presenting manifestation of occult malignancy. The diagnosis of hypertrophic osteoarthropathy should cause an intensive search of cancer. Further associated diseases are discussed.

Along with the great strides that have been made towards understanding cancer, has come a realization of the complexity of molecular events that lead to malignancy. Proteomics-based approaches, which enable the quantitative investigation of both cellular protein expression levels and protein-protein interactions involved in signaling networks, promise to define the molecules controlling the processes involved in cancer.

TITLE: [Squamous cell carcinoma of the face: therapeutic approach]
PUBDATE: 19981101
Cutaneous squamous cell carcinoma is the most common metastatic skin cancer. The importance of early recognition and thorough treatment of premalignant lesions as well as the recognition of risk factors of the neoplasms that are most likely to metastasize must also be emphasized. A retrospective study of 126 patients, treated for primary squamous cell carcinoma of the face, was undertaken over a 7-year period. An attempt is made to define the major therapeutic modalities chosen, taking into consideration the specific anatomic location.
